FACS Core. FACS is a fundamental technology required in all aspects of stem cell research. FACS allows rapid and accurate characterization of stem cell populations as well as isolation of rare stem cells or differentiated cells from contaminating cell populations.

The core has four modern sorters, a BDFACS Aria III, BD Influx and two Sony MA900 and four cytometers, a ThermoFisher Attune with an Auto Sampler, two BD Canto II's and a BD FACSSymphony. Users also have access to an imaging cytometer, the Amnis ImageStream MKII. There is also a Meso QuickPlex SQ120 Multiplex electrochemiluminescence The Flow Cytometry Facility, located at UMass Medical School in Worcester, provides a wide variety of cytometry services for internal and external investigators with state-of-the-art sorting and analyzing capabilities. For analysis we currently have three 4-color FACSCaliburs, a 2 laser LSR II, a 4 laser LSRII and a 5 laser LSRII. Our cell sorters include a 3 laser Beckman Coulter MoFlo XDP hi Prof. Dr. med. The mission of the Flow Cytometry (FACS) Core is to provide researchers at CHLA and other institutions with the ability to perform multi-parameter analysis and cell sorting on single-cell suspensions to asses cell morphology, phenotype, and function through standard and custom applications in support of biomedical research. Dilutions, if necessary, should be made in FACS buffer. Incubate for at least 30 min at room temperature or 4°C in the dark. This step will require optimization. Wash the cells 3 times by centrifugation at 1500 rpm for 5 minutes and resuspend them in 200 μl to 1ml of ice cold FACS buffer*.
